The actress said she was "beyond excited" about presenting the ceremony in Madrid next month.
Kings of Leon, Ke$ha and Kid Rock have been added to the line-up, which also includes Katy Perry and Linkin Park.
Perry, who hosted the show in 2008 and 2009, has tied with Lady Gaga for the most nominations, with a total of five.
The singers will go head to head in four of the five categories they have been nominated in, including best pop, best female, best song and best video.

Lady Gaga was the big winner at the MTV Europe Music Awards - but UK artists left empty-handed.
The US singer, who had been nominated for five awards, won Best Song for her hit single Bad Romance, as well as Best Pop and Best Female.
She was unable to pick up her gongs in person at the Madrid ceremony on Sunday night, but thanked fans from the stage during a concert in Budapest.
The 24-year-old told MTV viewers: "I hate that I'm not there, but I'm working so hard."
Katy Perry, also nominated for five awards, took Best Video for her song California Gurls, while Justin Bieber won Best Male and Best Push Artist.
But although British nominees included Plan B, Ozzy Osbourne, Muse and Alexandra Burke, none succeeded in scooping a trophy.
The awards, held at Madrid's Caja Magica, were hosted by Desperate Housewives star Eva Longoria, who stunned audiences with 13 costume changes, including an array of glamorous mini-dresses, a pair of hot pants and an outfit shaped like a giant Spanish ham.
The awards opened in style with a medley from Shakira, and included performances from acts as diverse as Plan B, Kid Rock, Rihanna and Linkin Park.
Kesha won the Best Newcomer award, while Thirty Seconds to Mars thanked their devoted fan-base, known as 'the Echelon' after beating Muse and Kings of Leon to Best Rock.
Linkin Park beat Muse and Kings of Leon to the award for Best Live Act, while punk-rock act Paramore triumphed in the Best Alternative category. And Bon Jovi, who closed the ceremony with a medley of their hit songs including It's My Life and Shot Through the Heart, were honoured with the first MTV Global Icon Award.
Lady GaGa was the big winner at last night's 2010 MTV EMAs in Madrid, scooping three awards.
The US popstar, who didn't attend the ceremony, was awarded the Best Pop, Best Female and Best Song gongs.
Other victors on the night were Ke$ha, who won the Best New Act title, Justin Bieber, who triumphed in the Best Male and Best Push Artist categories and rock act Paramore, who earned the Best Alternative win.
Eva Longoria hosted the ceremony at the Caja Magica , which featured performances from Katy Perry, Kings Of Leon, Miley Cyrus, Linkin Park and Ke$ha. Earlier in the evening Kanye West surprised fans by collaborating with 30 Seconds To Mars at a pre-show gig, during which he performed 'Hurricane' and a brief rendition of 'Power'.
